The Louisiana purchase affects B.N.A colonies because this purchase was most likely very intimidating for B.N.A colonies on the other side of the unguarded border. The north already had problems with B.N.A because they supported the south during the civil war. That could give U.S ideas about attacking B.N.A, and if they ever tried it would be easy for them given the large borders between U.S and B.N.A colonies. The Louisiana purchase shows manifest destiny because if the US now has a bigger piece of land it will build on their belief of them being superior to others and it is adding on their expansion, making them more powerful.
